The evidence comes from a peer-reviewed neuroimaging study published in PLOS ONE titled β€œPlaying Super Mario 64 increases hippocampal grey matter in older adults.” Researchers followed adults aged 55–75 who had little to no prior video-game experience and randomly assigned them to one of three groups for 6 months: a 3D video-game group playing Super Mario 64, a music-training group (piano lessons), or a passive control group. Brain MRI scans taken before and after the intervention showed that only the video-game group experienced a significant increase in hippocampal grey matter, a brain region critical for memory formation and spatial navigation.

Importantly, the control group that did not learn a new skill showed the opposite effect: measurable hippocampal grey-matter loss over the same period, consistent with age-related brain atrophy. The music group showed growth in other brain regions linked to executive function but not the hippocampus, highlighting that the effect was specific to the spatial, navigation-heavy demands of 3D gaming. Participants in the gaming group also showed improvements in short-term memory performance, which correlated with the increase in hippocampal volume, reinforcing that the structural brain changes were functionally meaningfulβ€”not just cosmetic MRI findings.
