pH Stat titration in detergent soap manufacturing
Enzymes are added in soap making to enhance the cleaning process. Enzymes like lipase are added to detergents where their function is to remove fatty residues and stains. The activity of these enzymes is measured using pH stat titration. As the pH value is increased above or decreased below the optimum pH, the enzyme activity decreases.
Commercially, many auto titrators are equipped to carry out such titrations. The most familiar ones are the AUT-701 by TOADKK and the TIM 856 pH Stat by Radiometer. This titration method is normally based on the rate at which the lipase enzyme hydrolyzes tributyrin at pH 7.0 to form butyric acid. The butyric acid is then titrated with sodium hydroxide, and the consumption of sodium hydroxide is recorded as a function of time.
During the hydrolysis process, butyric acid (a free fatty acid) is released from triglycerol, with the enzyme lipase acting as a bio-catalyst. The activity of the enzyme lipase determines the consumption of sodium hydroxide (NaOH). The method of analysis includes sample incubation and endpoint alkaline titration of the released acids.
The pH-stat titrator instrument is commonly used for this purpose. Titration is carried out at 30 degrees Celsius, for which special cells are required to maintain the sample temperature. The Lipase Unit (LU) is defined as the amount of enzyme that releases 1 µmol of butyric acid per minute under the given standard conditions.

Importance of turbidity measurements and technology used
Turbidity measurements are important for some applications in the pharmaceutical industry, whether for a finished drug or the equipment used in production. Turbidity measurements are performed to assess the presence of contaminants and nano suspensions. For drugs like cephalosporin and carbapenem, which are used to treat bacterial infections, lab chemists conduct turbidity measurements. This process guides the team to take necessary corrective and preventive steps in production. The turbidity limit for these drugs is 6 NTU.
In many pharmaceutical industries, production equipment such as glass reactors and metal machine parts are thoroughly washed and cleaned with high-purity water. The turbidity and Total Organic Carbon (TOC) of this water are measured before and after cleaning to check for contaminants and cleanliness. Pharmaceutical companies proceed with production only if these parameters are within the prescribed limits. To achieve this, modern nephelometers are used to detect turbidity in such samples. These devices are designed according to two methods: ISO 7027 and EPA 180.1. Method ISO 7027 uses a red LED light source with an 860 nm filter, while EPA 180.1 uses a tungsten lamp with a 507 nm filter.
These nephelometers are specially equipped with four detectors, making them well-suited for detecting trace-level turbidity below 0.1 NTU. The technology employs an arrangement of four detectors around the sample cell: one detector at 180°, measuring direct transmittance of light passing through the sample cell, and the other three observing scattered light from the sample cell, arranged at 90°, 45° forward, and 45° backward. The detectors at 45° forward and 45° backward are particularly useful for checking trace-level suspensions in the sample.
Method ISO 7027 is found to be more reliable compared to method EPA 180.1, primarily due to the light source. A red LED emits uniform and stable light intensity compared to a tungsten lamp, where variations in light intensity are observed. Another advantage of the red LED light over the tungsten lamp is its longer lifespan. Some manufacturers of turbidity meters for the EPA 180.1 method use halogen gas-filled lamps to achieve somewhat uniform light intensity. To garner a commercial advantage, these instruments often include additional photometric features such as absorbance, transmittance, and concentration, making them two-in-one instruments that function as both a turbidity meter and a photometer.
