0. 오가노이드를 이용하면 발생생물학 연구, drug screening, 동반진단 등 많은 응용이 가능합니다. 하지만 살아있는 오가노이드를 염색 등 전처리 없이, 고해상도로 측정할 수 있는 기술은 없었습니다. small molecule로 염색을 해서 제한된 영상을 보거나, 또는 fixation, sectioning후 antibody staining을 해서 2D로 관찰하는 방식을 따르게 됩니다. 오가노이드 그대로 상태를 유지하면서 고해상도 분석이 불가능 한 것이지요.

1. Holotomography로 살아있는 소장 오가노이드의 고해상도 label-free 이미징과 분석 논문을 발표했습니다. IBS 구본경 단장님팀의 organoids 배양 기술, TOMOCUBE의 2세대 홀로토모그래피 HT-X1과 분석 S/W 개발, 그리고 Biomedical Optics Lab, KAIST 이만재 박사의 측정과 분석을 통해 결과물이 나왔습니다.

2. 고해상도로 살아있는 organoids를 관찰할 수 있게 되었지만, 새로운 challenges들이 생겨났습니다. 150 um 보다 두꺼운 오가노이드에서는 multiple scattering으로 인한 blurring 문제가 발생하는데, 이를 극복해야만 더 큰 오가노이드에도 적용할 수 있습니다. 그리고 holotomography영상을 보고 세포의 종류와 상태를 분석할 수 있다면 멋질 것 같습니다.

Asian Society of Digital Pathology - ASDP 에서 초청 발표가 있었습니다. Holotomography와 AI를 이용해서 3D histopathology를 구현하는 방법을 소개했습니다. 일본과 싱가폴 연구진들도 매우 훌륭한 연구 결과를 발표해서 큰 자극을 받았습니다. digital pathology는 필연적인 흐름이고, 이렇게 얻어지는 방대한 pathology 정보를 어떻게 AI과 결합해서 어떤 새로운 가치를 만들어 낼 것인가?가 핵심 화두가 되었습니다.
