🧠 The GSK2 and Alzheimer's Disease
Researchers at ETH Zurich have uncovered a new biological pathway that may help explain how Alzheimer's progresses and how it could potentially be slowed.
At the center of the discovery is GRK2, an enzyme that normally helps brain cells respond to stress. But in Alzheimer's, GRK2 can become inactivated and clump together into toxic aggregates. These aggregates accumulate on mitochondria, the cell's powerhouses, blocking their pores and starving neurons of the energy they need to survive.
The damage doesn't stop there. The team found that inactive GRK2 also accelerates the production of amyloid-beta, one of Alzheimer's hallmark proteins. That amyloid-beta places even more stress on neurons, triggering additional GRK2 inactivation and creating a self-perpetuating vicious cycle of degeneration.
To interrupt this loop, researchers developed Compound 10, a molecule designed to prevent GRK2 aggregation.
In mouse models, Compound 10:
🔬 Preserved mitochondrial function
🧠 Reduced amyloid-beta accumulation
⚡ Delayed nerve cell death
❤️ Improved heart function
🐭 Extended survival
Perhaps most remarkable is the timeline behind this work. The project began nearly 20 years ago using human brain tissue samples from Ain Shams University Hospital in Cairo. Because Alzheimer's is an age-related disease, many experiments required older mice, which meant each study took years to complete.
